Acidic dyes have auxochromatic groups such as hydroxyl, carboxyl, and sulfone groups that are negatively charged in solution. The salt composed of this pigment and Na, K, and Ca ions is alkaline in solution. It combines with the important components of positively charged cells, or infiltrates into the intercellular space to stain cytoplasm, red blood cells, muscle fibers, etc.
